Identification and characterization of stressed degradation products of moexipril using LC-ESI-MS/Q-TOF

Abstract
A gradient LC-MS method has been developed for the identification and structural characterization of stressed degradation products (DPs) of moexipril (MOX) using liquid chromatography electrospray ionization tandem mass spectrometry (LC- ESI-MS/MS). MOX was subjected to hydrolytic (acidic, alkaline and neutral), oxidation, photolytic and thermal stress conditions as per ICH guidelines Q1A(R2). The drug degraded under hydrolytic, oxidative and photolytic conditions, while it was stable to thermal conditions. In total, 5 DPs were formed and the chromatographic separation of the drug and its DPs was achieved on an Agilent XDB C-18 column (4.6 X 150 mm, 5�¼m) with a mobile phase consists of 20mM ammonium acetate (pH adjusted to 6.0 by using NaOH solution) and methanol in an gradient elution mode at a flow rate of 1.0 mL/min at 25 O C. To elucidate the structures of degradation products, fragmentation of the [M+H]+ ions of MOX and its degradation products was studied by using LC-MS/MS and MSn experiments combined with accurate mass measurements. The product ions of all the protonated degradation products were compared with the product ions of protonated MOX to assign most probable structures for the observed degradation products.

V. Jayathirtha Rao has obtained his PhD (Prof. V. Ramamurthy) in 1983 from Indian Institute of Science, Benguluru, India; Post Doctorate (Prof. R. S. H. Liu) 1983-1984 at University of Hawaii, Honolulu, USA and as Research Associate (Prof. Koji Nakanishi) 1985-1986 at Columbia University, New York, USA. He is Head and Chief Scientist of Crop Protection Chemicals Division of Indian Institute of Chemical Technology, Hyderabad, which is a premier R&D institute in India. He is a four time Alexander vonHumboldt Fellow - Germany. He is honored with Fellow of The Royal Society of Chemistry (London) in 2008. He is a Fellow of Andhra Pradesh Academy of Sciences (FAPAS) ? Hyderabad. He is recognized as a Top Tier Reviewer for ACS Journals. He has published over 190 publications (papers + patents + processes + Book Chapters + Book). He has 29 PhDs and 52 Master students for his credit.
