Neurodegeneration induced by pathogenic proteins depends on contributions from surrounding glia. P-65 in neurons and
astrocytes are critically involved in neuronal death induced by amyloid-beta (Aβ) peptides, which are widely presumed
to cause Alzheimer diseases. The main inflammatory players in AD are the glial cells which initiate the inflammatory response.
One of the earliest neuropathological changes in AD is the accumulation of astrocytes at sites of Aβ deposition to protect the
neurons from toxic damage. It is desirable to find methods of tipping the balance towards anti-inflammatory state. Here we
demonstrate that cannabinoids in astrocytes in a primary culture increase producing changes in the signalling of these cells.
We previously found that Aβ induces inflammatory mediators, such as IL-1β and TNF-α and transcripction factor as p-65.
Stimulation of astrocytes with cannabinoids increases the action of the PPAR receptors protecting astrocytes against Aβ-
induced inflammation. Results support that cannabinoids can have a neuro protecting effect in the treatment of AD.
